GeneProof PCR kits were developed in two versions providing for control of the process correctness and progress of the sample processing procedure. The kits referred to as ISEX provide for control of both the nucleic acid isolation efficiency and the PCR reaction progress, i.e. of the whole diagnostic process. The control principle varies according to the DNA or RNA that is to be detected.

 

ISEX

In case of virus RNA detection we use the natural presence of the human gene mRNA encoding GAPDH in serum and plasma Detection of this mRNA in the channel for HEX fluorophore detection indicates the following:

  • Good sample quality - mRNA is a fragile molecule and it quickly degrades when the recommended transportation and storage conditions are not complied with - this implicates the risk of degradation for the viral RNA contained in the sample. Detection of this mRNA in the sample therefore implicates compliance with the storage and transportation conditions and it provides control for false negative results due to the sample degradation
  • RNA extraction efficiency control - positive result of the mRNA GAPDH presence indicates efficient RNA extraction. It therefore provides control for false negative results due to inefficient RNA isolation
  • Reverse-transcription PCR efficiency control - positive result of the mRNA GAPDH presence indicates correct transcription from RNA to DNA and correct progress of the subsequent polymerase chain reaction (PCR). It therefore controls false negative results due to the ineffective reverse-transcription PCR and/or PCR reaction inhibition

The control principle varies according to the DNA or RNA that is to be detected.

Kits referred to as ISIN provide for the PCR reaction progress control (PCR inhibition) only. This version is available for DNA detection.

ISEX

Detekce DNA

ISEX versions of the GeneProof PCR kits contain Internal Standard as an independent component. Internal Standard is an artificial DNA construct prepared by means of genetic engineering methods. The Internal Standard is added into the sample before the DNA extraction and its detection in the HEX fluorophore channel indicates the following:

  • Efficient DNA extraction - detection of this construct demonstrates that other DNA, i.e. the DNA of the microorganism to be detected, has been successfully detected along with the Internal Standard. Internal Standard use therefore controls for false negative results due to inefficient DNA isolation
  • Correct progress of PCR amplification - detection of this construct demonstrates the correct progress of the PCR reaction and control of the eventual inhibition of the PCR amplification. The Internal Standard is amplified from the same primers as the microorganism to be detected (competitive PCR), therefore the amplification efficiency for both molecules is similar, providing for much more efficient inhibition control then duplex systems

ISIN

ISIN versions of the GeneProof PCR kits contain Internal Standard as a part of the MasterMix. Internal Standard is an artificial DNA construct prepared by means of genetic engineering methods and its detection in the HEX fluorophore channel indicates the correct progress of the PCR amplification. The Internal Standard is amplified from the same primers as the microorganism to be detected (competitive PCR), therefore the amplification efficiency for both molecules is similar, providing for much more efficient inhibition control then duplex systems.
